    Dynamics of semi-flexible polymer solutions in the highly entangled regime

    We present experimental evidence that the effective medium approximation (EMA), developed by D.C. Morse [Phys. Rev. E {\bf 63}, 031502, (2001)], provides the correct scaling law of the macroscopic plateau modulus G0ρ4/3Lp1/3G^{0}\propto\rho^{4/3}L^{-1/3}_{p} (where ρ\rho is the contour length per unit volume and LpL_{p} is the persistence length) of semi-flexible polymer solutions, in the highly entangled concentration regime. Competing theories, including a self-consistent binary collision approximation (BCA), have instead predicted G0ρ7/5Lp1/5G^{0}\propto\rho^{7/5}L^{-1/5}_{p}. We have tested both the EMA and BCA scaling predictions using actin filament (F-actin) solutions which permit experimental control of LpL_p independently of other parameters. A combination of passive video particle tracking microrheology and dynamic light scattering yields independent measurements of the elastic modulus GG and LpL_{p} respectively.     Validation of an ion selective electrode system for the analysis of serum fluoride ion

    A high impedance unit was developed for use with a fluoride/pH electrode system for the measurement of serum fluoride. The linearity, accuracy, precision and detection limit of the system is reported. At a pH of 1.55, the system was linear over a range of serum fluoride concentrations up to 100 μmol l-1, with a lower limit of detection of 0.3 μmol l-1. Recoveries at this pH were 94-105% in the range 2.6-100 μmol l-1. Within-run CVs ranged from 4.2% at a level of 2.3 μmol l-1 to 1.2% at a level of 55.7 μmol l-1, while day-to-day CVs ranged from 12.8% at a level of 2.2 μmol l-1 to 4.6% at a level of 51.7 μmol l-1. The system demonstrated a rapid response time and has the potential for a smaller sample size requirement with alternative electrode shape. Continued development of this unit into an automated fluoride ion selective electrode system is recommended, since the measurement of serial serum fluoride samples is of greatest importance in assessing the impact of new anaesthetic agents on renal function

    The prepower stroke conformation of myosin V

    eW have used electron microscopy and single-particle image processing to study head conformation in myosin V molecules. We find that in the presence of ATP, many heads have a sharply angled conformation that is rare in its absence. The sharply angled conformation is similar to a myosin II atomic structure proposed to mimic the prepower stroke state. The leading head in molecules attached to actin by both heads has a similar conformation, but is also sharply angled in a second plane by tethering through the trail head. The lead head lever joins the motor domain ∼5 nm axially from where it joins the trail motor. These positions locate the converter subdomain and show the lead motor is in the prepower stroke conformation. Tethering by the trail head places the lead head motor domain at the correct axial position along the actin for binding, but at the wrong orientation. Attachment is achieved either by bending the lead head lever throughout its length or at the pliant point. The microscopy shows that most of the walking stride is produced by changes in lever angle brought about by converter movement, but is augmented by distortion produced by thermal energy

    Explaining motivation in language learning: a framework for evaluation and research

    Researching motivation in language learning is complex and multi-faceted. Various models of learner motivation have been proposed in the literature, but no one model supplies a complex and coherent framework for investigating a range of motivational characteristics. Building on previous models I propose such a methodological framework, based on a complex dynamic systems perspective, which re-conceptualises the investigation of motivation in SLA in qualitative and mixed method approaches by offering one flexible tool for case study approaches. This new framework has been tried and tested in three locations in England and reported as case studies. The study aimed to address the following research questions: (1) in what ways does CLIL impact on learner motivation? (2) what are the main elements of CLIL that enhance motivation? Overall analysis of the results found that where expectations of success were high and where the teaching was effective, CLIL had a positive impact on motivation and progress. The framework is designed to be flexible enough to be used to investigate language learning in a range of national contexts. It is hoped that the proposed framework, reported here together with exemplification and commentary from the English study, will enable researchers in a wide range of language learning contexts to investigate learner motivation in a systematic and in-depth manner

    Post-construction thermal testing: Some recent measurements

    In the UK, it has become apparent in recent years that there is often a discrepancy between the steady-state predicted and the measured in situ thermal performance of the building fabric, with the measured in situ performance being greater than that predicted. This discrepancy or gap in the thermal performance of the building fabric is commonly referred to as the building fabric 'performance gap'. This paper presents the results and key messages obtained from undertaking a whole-building heat loss test (a coheating test) on seven new-build dwellings as part of the Technology Strategy Board's Building Performance Evaluation Programme. While the total number of dwellings involved in the work reported here is small, the results illustrate that a wide range of discrepancies in thermal performance was measured for the tested dwellings. Despite this, the results also indicate that it is possible to construct dwellings where the building fabric performs thermally more or less as predicted, thus effectively bridging the traditional building fabric performance gap that exists in mainstream housing in the UK

    Role of Symbiotic Auxotrophy in the Rhizobium-Legume Symbioses

    Symbiotic auxotrophy occurs in both determinate pea and indeterminate bean nodules demonstrating its importance for bacteroid formation and nodule function in legumes with different developmental programmes. However, only small quantities of branched chain amino acids are needed and symbiotic auxotrophy did not occur in the Sinorhizobium meliloti-alfalfa symbiosis under the conditions measured. The contrasting symbiotic phenotypes of aap bra mutants inoculated on different legumes probably reflects altered timing of amino acid availability, development of symbiotic auxotrophy and nodule developmental programmes
